A triple receptor agonist activating GLP-1, GIP and glucagon receptors simultaneously. The most mechanistically complex incretin compound under active investigation. Phase 2 TRIUMPH data showed 24 percent mean weight reduction at the highest dose.
GLP-1R for appetite suppression and insulin secretion. GIPR for insulin sensitisation and adipose lipid metabolism. Glucagon receptor increases basal metabolic rate, promotes hepatic fat oxidation and activates thermogenic brown adipose tissue.
Each generation hits one more receptor. Semaglutide, one receptor: eat less. Tirzepatide, two receptors: eat less and handle insulin better. Retatrutide, three: eat less, handle insulin better AND burn more calories at rest. Phase 2 showed 24 percent average weight loss. Nothing approved today comes close.
